Macrophage Differentiation of Human Precursor B-Cell Line by Phorbol Ester and Colony-Stimulating Factor

摘要: A human round cell line, KLM-2, is considered to be precursor B-cell by immunologic surface marker analysis and histochemical studies. The effect of 12-O-tetradecanoyl phorbol-13-acetate (TPA) and/or colony- stimulating factor (CSF) on KLM-2 cells was investigated. became adherent the bottom flask day 2 after liquid culture with TPA, number macrophage-like (mAC) pseudopodia showed a TPA dose-dependent increase peak level 3 or 4. mAC had phagocytic activity. suspending were positive for alpha-naphthyl acetate esterase (NAE) up about 50% in TPA. concentration 100 ng/ml optimal provide maximum NAE-positive cells. Particularly, such effects markedly accelerated presence CSF. These findings indicate macrophage differentiation may bipotential stem line differentiating into monocyte-macrophage lineages.
