摘要: Linear density gradients of Renografin have resolved two components bacterial deoxyribonucleic acid (DNA) in sheared lysates. Component 1, at equilibrium after 5 hr centrifugation, is enriched for newly synthesized DNA and markers near the origin terminus replication. It contains 5% total cellular protein, 25% phospholipids, 30 to 50% DNA, 4 11% unstable ribonucleic (RNA), RNA polymerase, low amounts polymerase. The material sensitive Pronase Sarkosyl. In unsheared lysates, all forms a band this position. Shearing lysate generates slow-sedimenting fraction (component 2) which more uniformly labeled than DNA.
