摘要: A flow-cytofluorometric method, based on the differential stability of deoxyribonucleic acid versus ribonucleic with metachromatic dye, acridine orange, simultaneously measures following parameters stimulation in mixed lymphocyte cultures: (a) number nonstimulated cells; (b) total stimulated lymphocytes; (c) lymphocytes G1, S and G2 + M phases cell cycle; (d) macrophages; (e) dead cells. The progress may also be measured by a parameter representing accumulation per cell. method is rapid, avoids rinsing, fixation centrifugation applicable to microcultures. Multiparameter analysis which provides simultaneous measurements proliferation prove more sensitive assay histocompatibility than tests only (tritiated thymidine incorporation).
