摘要: Abstract Lymphocyte blast transformation in response to stimulation by phytohemagglutinin (PHA) or allogeneic cells one-way mixed lymphocyte culture has been quantitated using acridine orange (AO) staining and flow cytofluorometry. The method involves of fixed with AO such a manner that RNA stains metachromatically red DNA green. “Pyroninophillic” increased cytoplasmic are identified their high fluorescence per cell can be counted cytofluorometry stimulated unstimulated cultures. A comparison was made the proliferative normal peripheral blood lymphocytes (PBL) as determined incorporation [14C]thymidine into parallel microcultures. two methods show similar varying concentrations PHA increasing strength interaction. separately simultaneously measure increase number (proliferative response) per-cell synthesis (nonproliferative immunologic stimulation.
