Positron Emission Tomography Using [18F]Galacto-RGD Identifies the Level of Integrin αvβ3 Expression in Man

摘要: Purpose: The integrin α v β 3 plays a key role in angiogenesis and tumor cell metastasis is therefore an important target for new therapeutic diagnostic strategies. We have developed [ 18 F]Galacto-RGD, highly -selective tracer positron emission tomography (PET). Here, we show, man, that the intensity of F]Galacto-RGD uptake correlates with expression. Experimental Design: Nineteen patients solid tumors (musculoskeletal system, n = 10; melanoma, 4; head neck cancer, 2; gliobastoma, breast 1) were examined PET using before surgical removal lesions. Snap-frozen specimens ( 26) collected from representative areas low intense standardized values (SUV) F]Galacto-RGD. Immunohistochemistry was done -specific antibody LM609. Intensity staining (graded on four-point scale) microvessel density -positive vessels determined correlated SUV tumor/blood ratios (T/B). Results: Two showed no (mean SUV, 0.5 ± 0.1). All other accumulation SUVs ranging 1.2 to 10.0 (mean, 3.8 2.3; T/B, 3.4 2.2; tumor/muscle ratio, 7.7 5.4). correlation T/B immunohistochemical (Spearman9s r 0.92; P 0.84; expression normal tissue (benign lymph nodes, muscle) two without uptake. Conclusions: Molecular imaging humans as by immunohistochemistry. might be used marker individualized planning strategies -targeted drugs.
