摘要: Although fibroblasts play an essential part during the wound healing response, mechanisms by which they mediate tissue remodelling and contraction are still unclear. Using live cell matrix imaging within 3D free-floating fibroblast-populated collagen lattices as a model for contraction, we compared behaviour of range with low high abilities analysed effect broad spectrum MMP-inhibitor GM6001 on contraction. We identified two underlying one via direct cell-mediated contractile activity, second through degradation. These appear to be linked morphology regulated concentration matrix. Cells rounded proliferated in but did not remodel it efficiently, resulting poor ability contract matrices. elongated showed higher levels protrusive leading efficient inhibited week-long various extents different lines. However, quantitative analysis activity that consistently decreased dynamics about 20%, this was correlated significant reduction early Overall our results suggest although fibroblast-mediated depends both MMP-mediated degradation, efficiency treatment preventing might dynamics.
