摘要: Abstract Inhibition of gastric alcohol dehydrogenase (ADH) has been shown to enhance absorption in man under certain circumstances. To determine whether various medications might affect absorption, we screened 18 compounds for an effect on ADH. Salicylic acid, acetaminophen, propranolol, ethacrynic and three H2-receptor antagonists all inhibited rat ADH vitro, indicating that several commonly used have the potential absorption. Of these, cimetidine, ranitidine, nizatidine were studied further define their assess clinical relevance effect. Both ranitidine enhanced small doses (0.15 g/kg) morning by 63% 64% increased Cmax 48% 54% respectively (p less than 0.001), effects similar those reported others cimetidine. The given before dinner was greatly attenuated with increases 8% (NS) AUC 21% = 0.02). Cimetidine 800 mg hs did not 0.15 g/kg evening, cimetidine 400 bid decreased 14% 0.11). 300 qid had no larger at dinner. We conclude many ADH, but increase actual amount absorbed is quite small, demonstrable only special conditions.
