The analgesic effect of the GABA-agonist THIP in patients with chronic pain of malignant origin. A phase-1-2 study.

摘要: Fourteen patients with chronic pain of malignant origin were treated escalating doses THIP intramuscularly 5-30 mg in an open phase 1 study. Analgesic activity was demonstrated 60% the at level 20 and a dose response relation present. Side effects, sedation, dizziness, euphoria, nausea, blurred vision present up to 80% limiting. The maximum serum concentration reached within h after dosing 87% all administrations. Mean t1/2 1.52 +/- 0.63 clearance 0.49 0.181 min. Significant correlations between concentration, THIP, analgesic effect side effects. It is concluded that cannot be used for treatment cancer pain, not because insufficient but unacceptable
