摘要: Abstract Objectives To determine if combined administration of LIMK2 and JNK inhibitors in a rat model ED induced by cavernosal nerve (CN) injury could restore erectile function suppressing both apoptosis fibrosis via rectification molecular pathways related to the structural alterations. Methods Sixty 12-week-old male Sprague-Dawley rats were categorized into four groups: (i) Sham-surgery (Sham) group, (ii) CN-crush-injury (CNCI), (iii) CNCI group (CNCI+L+1.0J) treated with combination 10.0 mg/kg LIMK2-inhibitors low-dose (1.0 mg/kg) JNK-inhibitors, (iv) (CNCI+L+10.0J) high dose (10.0 JNK-inhibitors. Ten days after surgery, response, histological-studies, Western-blot was investigated. Results The showed reduced maximal ICP/MAP or AUC/MAP, decreased immunohistochemical-staining α-SMA, SM/collagen ratio, increased phospho-cJun-positive apoptotic cells, phospho-LIMK2-positive fibroblasts, cJun-phosphorylation, LIMK2/Cofilin-phosphorylation, Bcl-2/Bax protein-expression fibronectin, compared Sham group. Both CNCI+L+1.0J CNCI+L+10.0J groups improvements erectile-responses, content number ratio cJun phosphorylation. Their tendency be greater than those Also, two treatment groups, fibronectin observed. Conclusions This study suggests that inhibition may improve restoration cJun/Bcl-2/Bax LIMK2/Cofilin at ten CN injury.
