摘要: A potential approach to activate tumor-specific T cells is use live bacterial vectors deliver appropriate antigens in a highly immunostimulatory context. We constructed recombinant strain of Listeria monocytogenes (rLM) expressing murine tyrosinase-related protein-2 (TRP-2), nonmutated melanocyte-derived differentiation antigen expressed melanomas. Immunization C57Bl/6 mice with this rLM efficiently primed CD8 recognize the MHC class I-restricted TRP-2180-188 epitope and express IFN-gamma upon vitro peptide stimulation. Peptide-loaded target were lysed by TRP-2-specific cytotoxicity assays, immunized boosted TRP-2 functionally protected from subcutaneous challenge B16 melanoma cells. These results identify characterize anti-"self" T-cell responses induced L. an endogenous, tumor antigen.
