摘要: The lipoaspirate fluid (LAF) is emerging as a potentially valuable source in regenerative medicine. In particular, our group recently demonstrated that it able to exert osteoinductive properties vitro. This original observation stimulated the investigation of proteomic component LAF, by means LC-ESI-LTQ-Orbitrap-MS top-down/bottom-up integrated approach, which represents object present study. Top-down analyses required optimization sample pretreatment procedures enable correct intact proteome. Bottom-up have been directly applied untreated samples after monodimensional SDS-PAGE separation. analysis acid-soluble fraction LAF top-down approach allowed demonstrating presence albumin and hemoglobin fragments (i.e. VV- LVV-hemorphin-7), thymosins β4 β10 peptides, ubiquitin acyl-CoA binding protein; adipogenesis regulatory factor, perilipin-1 fragments, S100A6, along with their PTMs. Part bottom-up profile was reproducibly found both tested samples. proteins, listed among components adipose tissue and/or comprised within ASCs intracellular content secreted Our data provide first glance on molecular profile, consistent its environment. appeared contain bioactive peptides paracrine factors, suggesting potential translational exploitation.
