摘要: Longan (Dimocarpus longan L.) is a tropical/subtropical fruit tree of significant economic importance in Southeast Asia. However, lack transcriptomic and genomic information hinders research on traits, such as the control flowering. In this study, high-throughput RNA sequencing (RNA-Seq) was used to investigate differentially expressed genes between unique cultivar ‘Sijimi’(S) which flowers throughout year more typical ‘Lidongben’(L) only once season, with aim identifying candidate associated continuous 36,527 40,982 unigenes were obtained by de novo assembly clean reads from cDNA libraries L S cultivars. Additionally 40,513 assembled combined these libraries. A total 32,475 annotated BLAST search NCBI non-redundant protein (NR), Swiss-Prot, Clusters Orthologous Groups (COGs) Kyoto Encyclopedia Genes Genomes (KEGG) databases. Of these, almost fifteen thousand identified significantly (DEGs) using Reads Per kb per Million (RPKM) method. 6,415 DEGs mapped 128 KEGG pathways, 8,743 assigned 54 Gene Ontology categories. After blasting public sequence databases, 539 potential flowering-related identified. addition, 107 flowering-time longan, their expression levels two samples compared RPKM method, 15 confirmed real-time quantitative PCR. Our results suggest homologues SHORT VEGETATIVE PHASE (SVP), GIGANTEA (GI), F-BOX 1 (FKF1) EARLY FLOWERING 4 (ELF4) may be involved flowering trait ELF4 key gene. The identification related will provide new insight into molecular process regulating time woody plants.
