摘要: Purpose: Amplification and/or overexpression of HER2/neu have been documented in many types epithelial tumor, and evaluation is now gaining importance, because this mechanisms disease can be inhibited vivo using humanized monoclonal antibodies. The main purpose our investigation includes the prevalence alterations non-small cell lung cancer (NSCLC) at different molecular levels. Experimental Design: We performed a comprehensive series 115 NSCLC, fluorescence situ hybridization (FISH), real time reverse transcription (RT)-PCR, immunohistochemistry. Results: immunoreactivity was detected 26 specimens (23%), with 5 carcinomas (4%) showing intense staining. Real RT-PCR demonstrated mRNA all samples analyzed, levels above normal 54 (47%). FISH gene amplification 9 41 (22%). Conclusions: These results demonstrate that occur albeit significantly depending on technical assay used for assessment. It therefore likely inhibitory antibodies will appropriate treatment subgroup NSCLC patients. suggest other unrelated to could responsible or protein overexpression. FISH, RT-PCR, immunohistochemistry are complementary techniques activation, useful identification patients treated. very sensitive requires minimal amounts tissue testing, additional studies should evaluate its clinical application patient evaluation.
