摘要: Background and Purpose Antiplatelet agents are widely used for the prevention of ischemic stroke. However, their effects on thrombus formation have rarely been evaluated in experimental animals vivo. We introduce methods evaluating antithrombotic action gerbils several antiplatelet formation. Methods In 8 to 10 weeks age, we tightly compressed unilateral common carotid artery 2 minutes using device prepared this purpose damage endothelium. Thrombus damaged was observed directly through microscope 30 minutes. six animals, examined immediately after experiments by electron microscopy. studied antiplatelets injecting drugs intravenously before endothelial damage. Results control studies, 70% 90% developed thrombi arterial compression. The microscopic examination displayed association with platelet at site. Administration mg/kg aspirin, 3 ticlopidine, 0.3 1.0 ibudilast, a novel prostacyclin accelerator, decreased frequency significantly, whereas 20 aspirin dipyridamole failed decrease formation. Conclusions This model is considered useful because its feasibility high reproducibility. present results support view that lower dose may prevent cerebral vascular accidents as efficiently higher aspirin.
