The human hepatocyte growth factor receptor gene: complete structural organization and promoter characterization.

摘要: The hepatocyte growth factor (HGF) receptor is a tyrosine kinase transmembrane protein encoded by the c-met proto-oncogene. Here, we have isolated and characterized human genomic DNA clones containing entire coding sequence of HGF receptor. gene spans approx. 120kb in length consists 21 exons interrupted 20 introns. Two alternative splice sites corresponding to known mRNA transcripts are located at exon 10 14, respectively. Structural analysis 5'-regulatory region reveals that promoter lacks TATA or CAAT elements but has an extremely high G-C content multiple Sp1 binding sites. By transfection series chimeric reporter constructs variable for chloramphenicol acetyltransferase, identified two positive, one negative regulatory dictate transcription renal epithelial mIMCD-3 cells nucleotide positions -2615 -1621, -223 -68, -1621 -1093, Moreover, deletion mutation markedly reduced activity cells, suggesting essential establishing constitutive expression gene. These data provide fundamental information on organization, as well origin different isoforms, should facilitate further studies transcriptional regulation its expression.
