摘要: The neural crest provides an excellent model system to study invasive cell migration, however it is still unclear how molecular mechanisms direct cells precise targets in a programmed manner. We investigate the role of potential guidance factor, neuropilin-1, and use functional knockdown assays, tissue transplantation vivo confocal time-lapse imaging analyze changes chick cranial migratory patterns. When neuropilin-1 function knocked down ovo, fail fully invade branchial arches, especially 2nd arch. Time-lapse shows that siRNA transfected stop collapse filopodia at arch entrances, but do not die. This phenotype autonomous. To test influence population pressure local environmental cues driving we isochronically transplanted small subpopulations DiI-labeled into host embryos ablated neighboring, premigratory cells. analysis reveals migrate narrow, directed streams. Interestingly, with reduction function, form segmental streams, suggesting initial migration invasion arches are separable processes.
