摘要: We examined the functional properties of CK beta-11/MIP-3 beta/ELC, a recently reported CC chemokine that specifically binds to receptor, EBI1/BLR2/CCR7. beta/ELC is distantly related other and CXC chemokines in primary amino acid sequence structure. Recombinant human expressed from mammalian cell system showed potent chemotactic activity for T cells B but not granulocytes monocytes. An optimal concentration attracted most input within 3 h, comparable with stromal derived factor 1 (SDF-1), highly efficacious chemokine. equally naive CD45RA+ memory type CD45RO+ cells. also strongly both CD4+ CD8+ cells, attraction was greater. more chemoattractant than MIP-1 alpha, known chemoattractant. induced actin polymerization lymphocytes, chemotaxis completely blocked by pertussis toxin showing its likely EBI1/BLR2/CCR7, coupled G(alpha i) protein. calcium mobilization which could be desensitized SDF-1, suggesting possible cross-regulation their signaling.
