摘要: Melanoma is an aggressive skin malignancy with a high mortality. Astrocyte elevated gene-1 (AEG-1), downstream target of Ras and c-Myc, has been implicated in the development multiple tumours, but its role melanoma remains unclear. In present study, AEG-1 was explored through silencing. Our results showed that silencing inhibited proliferation cells, induced cell cycle arrest, reduced levels cyclin A, B, D1, E, cyclin-dependent kinase 2. AEG-1silencing also apoptosis cells altered cleaved caspase-3, B-cell lymphoma-2 (Bcl-2) Bcl-2 associated X protein. Moreover, suppressed migration invasion expressions activities matrix metallopeptidase (MMP)-2 MMP-9, activation Wnt/β-catenin signalling pathway cells. Furthermore, vivo experiments revealed growth xenografts nude mice. summary, our study demonstrates oncogenic suggests may serve as potential therapeutic treatment melanoma.
