Liposomes injected intravenously into mice associate with liver mitochondria

摘要: Liposomes encapsulating uranyl acetate or ferritin were injected intravenously into mice. At periods of 20 min, 1 h and 4 post-injection, animals killed, livers excised. Transmission electron micrographs liver tissue showed association oligolamellar liposomes with mitochondria for each time period. an average one out ten was associated liposomes. In most cases, the clearly enclosed in a cytoplasmic vacuole. Phagocytosis by Kupffer cells as well fusion primary lysosomes inclusion secondary observed. No difference intracellular fate observed when lactosylceramide incorporated liposome bilayers, suggesting that differences biochemical studies are at level liposome-plasma membrane interaction. When containing hepatocytes isolated collagenase perfusion hour later, transmission EM revealed presence these cells, vacuoles cytoplasm mitochondria. A freeze-fracture-etching analysis excised min after injection ferritin, further supported observation associate liver.
