摘要: Real-time TaqMan® RT-PCR (TaqMan® RT-PCR) assays were developed to detect the viruses associated with Rugose wood complex of grapevines. The detected Rupestris stem pitting-associated virus in genus Foveavirus, Grapevine A, B and D Vitivirus. coat protein was found be most conserved gene within viral species, therefore, primers probes for designed from multiple alignment sequence various isolates each virus. Comparisons also made between conventional one step detection these using four-fold serial dilutions both purified RNA crude extract prepared grapevine tissue. Results showed that more sensitive could at 32- 256-fold higher extract, respectively, compared RT-PCR. use an internal control (18S rRNA) allowed comparison sample preparation protocols amplification efficiencies samples.
