Tanshinone II-A attenuates cardiac fibrosis and modulates collagen metabolism in rats with renovascular hypertension

摘要: The adaptive changes that develop in the pressure-overloaded left ventricular myocardium include cardiac hypertrophy and interstitial fibrosis. objectives of present study were to evaluate effects Tanshinone II-A, a bioactive diterpene quinone isolated from Danshen, on fibrosis collagen metabolism rats with renovascular hypertension. Male Sprague-Dawley subjected two-kidney two-clip (2K2C) or sham operation (sham) treated Valsartan (Val, 26.7 mg/kg/d), II-A (Tsn, 70, 35 mg/kg/d) vehicle. Six weeks later, systolic blood pressure (BP), LV weight, abundance, function parameters, hydroxyproline content mRNA levels matrix metalloproteinase (MMP)-2, MMP-9, tissue inhibitor (TIMP)-1 TIMP-2 evaluated. Both high-dose (Tsn-H, 70 low-dose (Tsn-L, Tsn failed attenuate 2K2C-induced BP elevation but significantly attenuated attendant Val suppressed elevations (LVSP) 2K2C rats. Tsn-H exerted comparable suppressive gene expression MMP-9 TIMP-1, while decreased MMP-2 level without affecting transcript TIMP-2. dysfunction, Tsn-L showed slight improvement. These data demonstrate for first time, prevented improved rat model hypertensive independent hypotensive effect. conferred its beneficial probably through regulation MMPs/TIMPs balance.
