Anti-proliferative effects of Atractylis lancea (Thunb.) DC. via down-regulation of the c-myc/hTERT/telomerase pathway in Hep-G2 cells.

摘要: Atractylis lancea (Thunb.) DC. (AL), an important medicinal herb in Asia, has been shown to have anti-tumor effects on cancer cells, but the involved mechanisms are poorly understood. This study focused potential and molecular of AL proliferation Hep-G2 liver cell line vitro. Cell viability was assessed by MTT test cells incubated with ethanol extract AL. Then, apoptosis cycle progression were determined flow cytometry. Telomeric repeat amplification protocol (TRAP) assays performed investigate telomerase activity. The mRNA protein expression human reverse transcriptase (hTERT) c-myc real-time RT-PCR Western blotting. Our results show that effectively inhibits a concentrationand time-dependent manner. When treated after 48h,the IC 50 about 72.1 μg/ mL. Apoptosis induced via arresting G1 phase. Furthermore, reduced activity through inhibition hTERT c-myc. Hence, these data demonstrate exerts anti-proliferative down-regulation c-myc/hTERT/ pathway.
