摘要: We investigated the feasibility of using genetic techniques to census pine marten (Martes martes) populations by genotyping non-invasively collected samples (plucked hair and scats), with particular reference genetically depauperate Irish population. Novel real-time polymerase chain reaction methods were developed for species sex identification, targeting short DNA sequences. Background variation at 17 microsatellite loci was very low in population, an average 2.29 alleles per locus expected heterozygosity 0.35. Despite such polymorphism, a panel eight sibling probability identity 0.011 reliably identified individual their gender, as determined genotypes live trapped individuals. With high nuclear amplification success rates (93.8%) error (1.8%), plucked hairs may represent more reliable cost-effective source than scats monitoring this elusive carnivore, similar taxa sympatric stone Martes foina.
