摘要: A highly sensitive sandwich immunoassay of macromolecular antigens using the rabbit antibody Fab′—β-d-galactosidase complex and immunoglobulin-G-coupled glass rod as a solid phase is described. The Fab′ fragments IgG are conjugated with β-d-galactosidase from Escherichia coli N,N′-o-phenylenedimaleimide. Rabbit coupled to aminoalkylsilyl rods (3 mm in diameter 5 length) glutaraldehyde. wide range concentrations fraction (20–2000 μg/ml) effective for coupling, amount immunoglobulin G can be controlled. smallest amounts ornithine σ-aminotransferase rat liver, human 2,4-dinitrophenyl that determined 0.03, 0.3 0.04 fmol, respectively. sensitivity assay these affected mainly by non-specific binding complexes ability antigen molecules, adsorbed on phase, bind specifically complexes. simpler more than Sepharose 4B.
