摘要: A repetitive sequence in Xanthomonas campestris pv. juglandis was identified as a new insertion sequence, IS1403, by nucleotide analysis. homologous IS, IS1404, also cloned from X. campestris. Both shared at least 91% identity on level. The sequences had 42-bp terminal inverted repeat with eleven mismatches, and were flanked 4-bp target site duplications. Comparison of sites flank- ing IS1403 IS1404 revealed no similarities, the G+C content flanking 45%, but that 74%, indicating elements transpose quite randomly do not prefer A-T-rich regions. contained two GTG-initiated open reading frames (orfA orfB), orfB phase -1 relative to orfA. There an 6 G motif followed potential stem-loop structure between frames, which may have promoted translational frameshift produce transframe protein. deduced amino acid orfA α-helix-turn-α-helix DNA-binding D,D(35)E domain transposases, respectively. Based features discussed, placed among IS3 family IS407 Pseudomonas cepacia, IS476 vesicatoria, IS1222 Enterobacter agglomerans, ISR1 Rhizobium lupini. widely distributed existed multiple copies most pathovars campestris, other plant pathogenic bacteria, such P. syringae apii, tabaci, glycinea, coronafaciens, Erwinia amylovora, E. carotovora, cypripedii, did contain this based Southern hybridization tests.
