摘要: 1. A human glioma cell line, NG97, was established by Grippo et al. in 2001 from tissue obtained a grade III astrocytoma (WHO, 2000). In this first study, the line grew as two morphologically distinct subpopulations: dendritic/spindle cells and small round cells. The injection of NG97 into nude mice induced an aggressive tumor characterized by: severe cytological atypia, vascular proliferation pseudopalisading necrosis (glioblastoma multiforme features). 2. The purpose present study to characterize immunophenotype ultrastructural aspects using parental tumor, cultured xenotransplant, order assess its glial nature possible divergent differentiation. 3. NG97 xenotransplant expressed main neuroglial markers (GFAP, S-100 protein, NSE Leu-7) showed no aberrant expression other histogenetic markers. GFAP similarly culture, but decreased xenotransplant. reduced cells, substantially recovered This variability interpreted either phenomenon dedifferentiation or microenvironmental selection specific subclones. equally three contexts. xenotransplant’s features were those highly undifferentiated tumor. No significant immunophenotypic differences between populations found. 4. Thus, our data demonstrate that constitute pure glial-committed which may prove useful malignant model studies addressing pathophysiological, diagnostic therapeutic issues.
