摘要: PURPOSE. Sub-RPE deposits form in a variety of conditions most notably age-related macular degeneration. The purpose this study was to generate sub-RPE vitro and test the hypotheses that high protein concentrations or retinal homogenate increase deposit formation challenge with tumor necrosis factor (TNF)-alpha metalloproteinase (MMP)-2 decreases such deposits.METHODS. ARPE-19 cells were grown on plastic collagen type I- coated membrane inserts media containing various fetal calf serum (FCS), bovine albumin, porcine homogenate. In addition, treated TNF-alpha MMP-2. assessed by electron microscopy classified into fibrillar, condensed, banded, membranous subtypes. area micrograph occupied each estimated point-counting technique. MMP-2 activity tissue culture supernatants zymography.RESULTS. With increasing time culture, total did not change, but amount condensed material deposited increased while fibrillar component decreased. Albumin resulted an deposit, predominantly type. Challenge led greater net significant increases banded forms. Cells showed dramatic reduction all types deposit. Zymography demonstrated unchallenged produced Retinal reduced active produced, stimulated MMP-9 production.CONCLUSIONS. formed share ultrastructural features those seen vivo. Deposit can be modulated homogenate, TNF-alpha, Significantly, results provide proof principle modified vitro.
