Effect of sucrose on shoot regeneration in Agrobacterium transformed Hypericum perforatum L. roots

摘要: The aim of the present study was to investigate effect sucrose on shoot regeneration potential in Hypericum perforatum L. roots obtained by Agrobacterium rhizogenes transformation. morphological evaluation transgenic grown media supplemented with (0.5, 1, 2, 4, 6 and 8 %) indicated that both genotype concentration significantly affected root elongation branching, as well regeneration. For two five analyzed clones, lower concentrations (up 2 %) led intensive regeneration, while other three clones intensified development only at elevated (4 %). all above 4 % had a deleterious development. Genetic characterization regenerated shoots revealed tested were diploid an average 0.670 ± 0.002 pg DNA per nucleus, no significant differences between non-transformed plants and, according PCR, integrated A. rolA, -B, -C -D genes. Real-time RT-PCR confirmed expression -B -C, rolD gene not detected. Differences detected absolute amounts transcripts highest levels for rol genes clone previously defined having high differentiation less effective potential. observed variations morphogenesis could be attributed different genes; additionally pointed out these trends.
