摘要: Abstract Purpose Develop an autologous culture method for ex vivo expansion of human limbal epithelial progenitor cells (LEPCs) using Plasma Rich in Growth Factors (PRGF) as a growth supplement and scaffold the LEPCs. Methods LEPCs were cultivated different media supplemented with 10% fetal bovine serum (FBS) or PRGF. The outgrowths, total number cells, colony forming efficiency (CFE), morphology immunocytochemistry against p63- α cytokeratins 3 12 (CK3-CK12) analyzed. PRGF was also used to elaborate fibrin membrane. effects on preservation stemness phenotypic characterization investigated through analysis CK3-CK12, ABCG-2 p63. Results showed significantly higher area than FBS cultures. Moreover, FBS, while displaying better overall. CFE found be groups compared p63-α expression differed between groups. membranes appeared confluent monolayer still retained p63 expression, being negative CK3-CK12. Conclusions can corneal tissue engineering, supplementing media, even basal without any other additives, well providing culture.
