Identification of anthranilate and benzoate metabolic operons of Pseudomonas fluorescens and functional characterization of their promoter regions

摘要: In an effort to identify alternate recombinant gene expression systems in Pseudomonas fluorescens, we identified genes encoding two native metabolic pathways that were inducible with inexpensive compounds: the anthranilate operon (antABC) and benzoate (benABCD). The antABC benABCD operons by homology Acinetobacter sp. putida operon, confirmed be regulated or benzoate, respectively. Fusions of putative promoter regions E. coli lacZ constructed confirm expression. Each was found controlled AraC family transcriptional activator, located immediately upstream first structural each respective (antR benR). We have promoters useful for tightly controlling at both small (< 1 L) large (20 fermentation scales.
