摘要: Abstract The effect of SK&F 104353 (2-hydroxy-3-carboxyethyltio-3-[2-(8-phenyloctyl)phenyl] propanoic acid), a specific leukotriene D4 receptor blocker, on volume regulation in isolated rat colonic crypts was studied. (10−7−5 × 10−6 mol · 1 −1) concentration dependently inhibited the regulatory decrease following cell swelling induced either by uptake short-chain fatty acid, butyrate, or exposure to hypotonic medium. Whole-cell patch-clamp experiments revealed that suppressed depolarization crypt cells during volume. Also (5 10−7 1−1), which has an action membrane potential similar swelling, nearly completely blocker. In contrast, precursor D4, C4, had no potential. These results are accordance with hypothesis acts as mediator for activation basolateral Cl− channels epithelium
