摘要: The zwitterionic detergent CHAPS, a derivative of the bile salts, is widely used in membrane protein solubilization. It "facial" detergent, having hydrophilic side and hydrophobic back. objective this work to characterize interaction CHAPS with cell membrane. To aim, erythrocytes were incubated wide range concentrations order determine partition behavior, its effects on lipid order, hemolytic effects, solubilization phospholipids cholesterol. results compared those obtained nonionic Triton X-100. was found that has low affinity for erythrocyte (partition coefficient K=0.06mM(-1)), at sub-hemolytic it causes little effect order. hemolysis phospholipid are closely correlated. On other side, binding X-100 disorders all levels, independent mechanisms Differential behavior observed Thus, resistant membranes (DRM) two detergents will have different composition. behaviors related differences their molecular structures, suggesting does not penetrate bilayer but binds flat position surface, both intact cholesterol depleted erythrocytes. A relevant result directly correlated lipids, as usually assumed.
