Haloperidol given chronically decreases basal dopamine in the prefrontal cortex more than the striatum or nucleus accumbens as simultaneously measured by microdialysis

摘要: Simultaneous microdialysis was performed in the prefrontal cortex, striatum, and caudal region of nucleus accumbens using implanted guide shafts with removable probes. Extracellular dopamine (DA), dihydroxyphenylacetic acid (DOPAC), homovanillic (HVA) were measured response to acute chronic haloperidol, an antipsychotic drug. Six rats received haloperidol (0.5 mg/kg, IP) while changes DA its metabolites monitored for six hours. Then probes removed daily injections given 28 more days. Next reinserted, a final challenge given. A control group treated same way but saline. The first injection increased extracellular DA, DOPAC HVA proving turnover all three regions. month later, after treatment, basal decreased just cortex. also cortex suggesting decrease at these two sites. None occurred accumbens. last treatment no longer any sites, production still detectable. This suggests lack responsiveness In summary, affected each regions differently; STR PFC; PFC.
