摘要: Human induced pluripotent stem cells (hiPSCs) are invaluable resources for producing high-quality differentiated in unlimited quantities both basic research and clinical use. They particularly useful studying human disease mechanisms vitro by making it possible to circumvent the ethical issues of embryonic cell research. However, significant limitations exist when using conventional flat culturing methods especially concerning expansion, differentiation efficiency, stability maintenance multicellular 3D structure establishment, prediction. Embryoid bodies (EBs), aggregates spontaneously generated from iPSCs suspension system, might help address these issues. Due unique microenvironment communication EB that a 2D culture system cannot achieve, EBs have been widely applied hiPSC-derived show advantages scaling up culturing, efficiency enhancement, ex vivo simulation, organoid establishment. can potentially also be used early prediction iPSC capability. To improve feasibility EB-mediated generate high quality EBs, critical factors including pluripotency maintenance, generation uniform morphology micro-pattern systems, proper cellular density inoculation, size control discussed on basis published data our own laboratory experiences.
