摘要: Previous work (Maxwell and Forbes: Development 101:767–776, 1987) has shown that an overlay of reconstituted basement membrane-like (RBM) gel dramatically increased the number catecholaminepositive (CA+) cells which differentiated in neural crest cultures. We report here this increase was inhibited when cultures were grown for 7 days presence agents elevate cAMP, such as 8-bromo-cAMP 3-isobutyl-1-methylxanthine. The action dose dependent with a half-maximal effect at about 50 μM. development CA+ reduced present from 0–4 vitro, but relatively unaffected if 4–7 vitro. tyrosine hydroxylase immunoreactive also by 8-bromo-cAMP. addition melanocytes resulted either no change or only modest reductions E/C8 neurofilament cells, indiciting on cell ontogeny selective. In contrast to effectof 8-bromo-cAMP, 8-bromo-cGMP did not inhibit RBM nor it stimulate absence overlay. Our results suggest cAMP may be important regulator phenotypic expression least some lineages. particular, these observations provide evidence levels can act prevent essential part normal cellular response leading appearance phenotype
