摘要: Abstract The responses of AMP-activated protein kinase (AMPK) and Ornithine decarboxylase (ODC) to isoproterenol have been examined in H9c2 cardiomyoblasts, AMPK represents the link between cell growth energy availability whereas ODC, key enzyme polyamine biosynthesis, is essential for all processes it thought a role development cardiac hypertrophy. Isoproterenol rapidly induced ODC activity cardiomyoblasts by promoting synthesis this effect was counteracted inhibitors PI3K/Akt pathway. increase became significant 15 30 min after treatment. At same time, stimulated phosphorylation AMPKα catalytic subunits (Thr172), that associated an acetyl coenzyme A carboxylase (Ser72) phosphorylation. Downregulation both α1 α2 isoforms subunit siRNA knockdown enzymatic activity, led superinduction isoproterenol-treated cardiomyoblasts. increased even cells treated with other adrenergic agonists control cells. Analogue results were obtained SH-SY5Y neuroblastoma transfected shRNA construct against AMPKα. In conclusion, quickly activates two events seem contrast one another. first one, linked growth, second, activation, homeostatic mechanism negatively modulates first. modulation may contribute processes.
