摘要: Background Vascular permeability is controlled by endothelial cell-to-cell junctions. (VE)-cadherin, the major component of adherence junctions (AJ) in endothelium, target permeability-increasing agent tumour necrosis factor-a (TNF-alpha). We investigated whether TNF-alpha regulates synthesis VE-cadherin on transcriptional level. Materials and methods Human cells, isolated from aorta (aEC) umbilical cord (HUVEC), were exposed to (200 U/ml) for 6 h or 12 h, with without subsequent incubation 24 h. mRNA was evaluated semi-quantitative RT-PCR. The VE-Cadherin protein expression analyzed flow cytometry (FACS). Results amplification curve treated cells shifted right compared controls, indicating a lower amount. number cycles at which half-maximal (N50) achieved, control aEC (30.7) than (33.0). N50 HUVEC + medium (N50 = 32.1), higher controls 29.7) 30.8). As determined FACS analysis, caused small decrease 7.44 6.05 mean channel intensity. Conclusion Our results indicate that affects gene level, inducing downregulation expression.
