摘要: Variation in the ovine DQA1 gene was investigated by amplification of exon 2 using PCR, followed single-strand conformational polymorphism (SSCP) analysis, cloning, and DNA sequencing. Fourteen novel SSCP patterns, representing 14 different sequences, were identified. Eight these sequences identical to published from sheep, whereas remaining six but similar sheep cattle. These new exhibited conserved region variable patterns than DQA2 sheep. All putative fulfilled criteria used established bovine leukocyte antigens major histocompatibility complex nomenclature committee for assignment as alleles. Comparison available cattle revealed several clusters some alleles more other The occurrence trans-species suggests action balancing selection at locus. Twenty-four percent nucleotide positions showed variation within 2, this seems have arisen largely point mutation conversion. nonsynonymous synonymous substitution rates both antigen-binding site codons nonantigen-binding codons. extensive reported article is consistent with
