摘要: Although cyclin E gene amplification is reported to be an important event in various cancers, it rarely found human colorectal cancers. As one of the candidate factors other mechanisms relating E, we analyzed E-dependent kinase activity cancer. Protein levels its catalytic subunit, cyclin-dependent 2 (Cdk2), and p21 p27 were determined by western blot or immunohistochemistry 27 cancers 10 adenomas, compared with adjacent normal colonic mucosa. Enzymatic E–Cdk2 complex neoplasm was measured using in-gel assay glutathione S-transferase-retinoblastoma (GST-Rb) fusion protein as substrate, that We clearly showed although level cancer adenoma similar mucosa, cylin increased all cases 90% adenomas. The relative significantly higher (3.7 ± 1.7 -fold) than adenomas (2.0 0.8-fold) (P < 0.004). expression Cdk2 (4.4 2.4 vs 2.7 1.3, P 0.04), not detected notably decreased adenoma. results this study strongly suggest activation may play role cancer, appears related amounts rather level.
