摘要: We previously reported a clinical study in which probucol reduced the restenosis rate. The mechanism of this effect is unclear. Restenosis characterized by neointimal hyperplasia caused proliferation smooth muscle cells (SMCs), increases expression Platelet-derived growth factor (PDGF)-A and SMemb. SMemb, non-muscle-type myosin heavy chain most predominantly expressed embryonic muscle, can be used as good molecular marker for dedifferentiated SMC. aim was to analyze on level PDGF-A SMemb after balloon injury rabbits. Probucol given orally 1.3 g/d from 2 weeks prior carotid time killing (2 or 4 injury). Intimal area determined histologically using computerized morphometry program. For quantification SMC proliferation, alpha-actin-positive proliferating cell nuclear antigen (PCNA)-labeled were counted. mRNA analyzed RNase protection assay. also examined immunohistochemistry. remarkably decreased intimal 70% number PCNA-labeled intima. significantly increased untreated rabbits, whereas it markedly suppressed with treatment. injured arteries at protein levels. However, did not suppress expression. effective preventing possibly due decrease PDGF.
