Analysis of cortisol, methylprednisolone, and methylprednisolone hemisuccinate

摘要: Abstract A sensitive, selective, and reproducible high-performance liquid chromatographic assay for the simultaneous measurement of cortisol methylprednisolone using dexamethasone as internal standard is presented. Samples are extracted with methylene chloride, washed sodium hydroxide then water, chromatographed on a microparticle silica gel column ultraviolet detection at 254 nm. Sensitivity greater than 10 ng/ml intra-day coefficient variation less 5% both steroids. The use porcine liver esterase allows quantitation hemisuccinate ester methylprednisolone. This has been applied in pharmacokinetic studies including investigations troleandomycin—methylprednisolone interactions. typical plasma concentration—time profile its prodrug presented one subject before after receiving troleandomycin therapy. Although elimination reduced presence therapy, there no effect pharmacokinetics succinate.
