摘要: Osteopontin (OPN) is an important matrix protein of renal calcium stone. However, the function OPN in early phase crystal formation not well defined. In this study, we examined expression with ultra-microstructural observations and immuno-TEM (transmission electron microscopy) control knock-out (OPN-KO) mice. Glyoxylate (100 mg/kg) was intra-abdominally administered to male wild-type mice (C57BL/6, 8 weeks age) OPN-KO age). Kidney collected before 6, 12, 24 h after administration. We relation between microstructural location using TEM immunohistochemical staining as western blotting quantitative RT-PCR for OPN. gradually increased cortex-medulla junction glyoxylate administration, mRNA until 12 h, but decreased at h. observation, began appear on luminal side distal tubular cells 6 detected lumen present nuclei collapsed mitochondria lumen. mice, were present, no Based results study proposed that appearance organelles, such microvilli, cell injury may be starting point nucleus due aggregation ability
