摘要: Background and Purpose: Previous efforts to identify the protein content of stone matrix have been limited by lack technology necessary analyze highly insoluble protein-crystalline complex. Our study objective is characterize calcium oxalate monohydrate (COM) stones using a comprehensive proteomics approach. Materials Methods: Seven pure COM were powdered, proteins extracted four different buffer solutions. Detergent cleanup spin columns or concentrators used remove detergent exchange buffers before trypsin digestion. Tryptic peptides analyzed with reversed-phase, high-performance liquid chromatography (RP-HPLC) tandem mass spectrometry (MS/MS) QSTAR Pulsar i quadrapole time flight spectrometer.
