摘要: Abstract A low-cost DNA sequencer was constructed based on a single helium-neon laser. The two-color peak-height encoded sequencing protocol, the use of T7 polymerase in manganese buffer, used to generate samples. Two termination reactions were performed. In first, TAMRA (applied Biosystems)-labeled primer extended presence ddATP and ddCTP. amounts dideoxynucleotides adjusted produce 3:1 peak height ratio. Similarly, ROX (Applied ddGTP ddTTP; pooled fragments separated 4% T LongRanger gel operated at 39°C. Over 500 bases sequence generated 50 min.
