摘要: In order to investigate the biological role of fibronectin in glioma cell invasion, we studied relation between migratory responses or adhesiveness cells and vitro invasion three human malignant lines, A172, T98G U373MG. All these lines chemotactically migrated a dose-dependent manner concentrations ranging from 0.5 10 microg/ml, with A172 showing strongest migration U373 weakest. Checkerboard analyses demonstrated that showed much stronger chemokinetic than U373MG cells. contrast responses, an almost equally high adhesion low adhesion. The degree expression integrin alpha5 subunit correlated well strength rather molecule. Furthermore, was completely inhibited by arginine-glycine-aspartic acid (RGD)-containing peptides, but fibronectin-stimulated only partially inhibited. An assay disclosed invaded artificial basement membrane barrier most least. However, addition markedly enhanced invasive activity had little effect on These results indicate can be one factors promoting invasiveness may play crucial through conferring motor-driving force
