摘要: The effect of caffeine when combined with cocaine or amphetamine was studied in rats. Animals were pretreated intraperitoneal vehicle (normal saline [NS]) 100 mg/kg, then challenged (0, 35, 50, 70, 90 mg/kg) d-amphetamine 15, 25, 42 mg/kg). Animal behavior, time to, and incidences seizures death recorded. This dose alone did not cause death. Caffeine pretreatment significantly incresed the incidence overt induced by either amphetamine. increased cocaine-induced from 10% to 90% at 70 mg/kg (P < .01). amphetamine-induced 0% 80% 15 ≤ .01), 70% 25 30% 35 To investigate mechanisms, additional animals withtthe adenosine agonist, 2-chloroadenosine (2.5 10 mg/kg), before being NS, cocaine, Pretreatment had no affect reducing toxicity. Combination potentiated phosphodiesterase inhibitor, pentoxifylline, potentiate authors conclude that potentiates acute toxicity both amphetamine, failure alter this suggests stimulants may be modulated nonspecific rather than specific adenosine- phosphodiesterase-induced mechanisms.
