Dietary Nucleotides: Effects on Cell Proliferation Following Partial Hepatectomy in Rats Fed NIH-31, AIN-76A, or Folate/Methyl-Deficient Diets

摘要: The requirement of a number tissues for dietary nucleotides could explain some the differences observed in animals fed natural ingredient diets vs. those purified lacking source nucleotides. Lack is exacerbated folate- or methyl-deficient semipurified diets, which both salvage and folate-dependent de novo synthetic pathways are diminished. We examined hepatocyte proliferation following partial hepatectomy weanling male Fischer-344 rats NIH-31 diet, nucleotide-free AIN-76A diet basal similar to but deficient methyl donors folate, choline methionine. Additional groups were folate/methyl-deficient supplemented with 0.25% yeast RNA. Compared NIH-31, increased dUMP/dTTP ratios, reduced mitotic index (MI) ratio proliferating cell (PCI) cells, an indication that hepatocytes delayed S-phase. Addition RNA reversed (by approximately 50%) effects on proliferation. A also produced markedly MI, increasing PCI/MI, suggested even further delay cells was effective significantly reversing folate/methyl deficiency.
