Voltammetric determination of trace quantities of 6-thioguanine based on the interaction with DNA at a mercury electrode

摘要: Abstract Herein, a sensitive square wave voltammetric (SWV) method is described for the quantitative determination of an anticancer drug, 6-thioguanine (6-TG). The interaction 6-TG with double stranded DNA (ds-DNA) in solution phase resulted well amplified SWV response at surface hanging mercury dropping electrode (HMDE). Accumulation and stripping steps were made sample medium conditioned acetate buffer (pH 4.8). Optimized conditions accumulation step included deposition potential −0.10 V, time 30 s, frequency 50 Hz, pulse amplitude 20 mV, 7 mV. In containing 2.0 mg L −1 ds-DNA, was performed within wide concentration range 2.4 × 10 −9 to 1.8 × 10 −5  mol L , detection limit 2.1 nmol L 6-TG. An overall conclusion that intercalation into ds-DNA possible reason observed behavior. applied tablets spiked blood serum samples. No statistically significant differences between expected obtained concentrations. new sufficiently detect ultra trace amounts content.
