Development of a highly reproducible three-dimensional organotypic model of the oral mucosa

摘要: In this report we describe the development of a standardized three-dimensional (3D) system human oral mucosa based on an immortalized keratinocyte cell line (OKF6/TERT-2). The procedure takes approximately 2-3 weeks to complete and includes three main stages: preparation collagen-embedded fibroblasts, addition mucosal component airlifting cultures ensure adequate differentiation/stratification. This results in multilayer epithelial structure which layers are organized similarly cells native mucosa. Specifically, model consists stratum basale, having one layer columnar round cells, relatively flattened spinosum granulosum, non-keratinizing corneum. 3D resembles commercially available TR146 (SkinEthic), with exception that our does not contain dyskeratotic changes has submucosal component, thus better represents normal submucosa.
