Do mucosal mast cells contribute to the immediate asthma response

摘要: In rat trachea, two types of mast cells have been identified, connective tissue (CTMCs) and mucosal (MMCs). We previously reported that CTMCs play an important role in tracheal contraction vitro via 5-hydroxytryptamine (5-HT) release a model. this study, we investigated whether MMCs also by employing cell-deficient (Ws/Ws) rats their congenic (+/+) rats. Rats were actively sensitized with ovalbumin challenged it 2 weeks later. To exclude the influence CTMCs, pretreated for 7 days compound 48/80 injected i.p. increasing doses. Histological study confirmed degranulation occurred but still remained. Histamine levels trachea decreased to 9.31% control levels. Ovalbumin-specific IgE production showed time-dependent increase both Ws/Ws +/+ after sensitization no significantly different values between groups. Ovalbumin challenge caused rats, not 48/80-pretreated Ketanserin inhibited contraction, leukotriene antagonist ONO-1078 did not, indicating was due 5-HT, whereas leukotriene, mediator specific derived from MMCs, has significant effect. The results suggest minimal, if any, contribution might another function. Furthermore, provide good model studying immunologic response airways.
